Protein Extraction

When extracting RNA, extend the grinding time relative to protein extraction protocols and include the RNA extraction buffer during the grinding process itself. The optimal duration depends on the tissue type and quantity. For tough samples, 2-3 grinding cycles may be necessary. Adjust all parameters based on experimental observations.

Grinding of strong and tough tissues

For resistant samples like bone or hair, processing in a 2 mL grinding tube is recommended. Mince the sample to 1-2 mm fragments, add grinding beads, and seal the tube. Snap-freeze in liquid nitrogen for 2-3 minutes. Perform an initial dry grind at 7 m/s for 60 seconds before adding the extraction solution. After adding the solution, grind again. If necessary, the freeze-grind cycle can be repeated to achieve complete homogenization.
